Abstract in Brazilian Portuguese: O uso da quimioterapia para o tratamento do câncer, mesmo com todo avanço alcançado, ainda apresenta, em alguns casos, fatores limitantes como invasividade, intensos efeitos colaterais e a não-responsividade, motivando assim, a contínua busca por novos agentes antitumorais. A classe química das quinoxalinas apresenta ampla gama de ações biológicas, dentre elas, atividade antitumoral. Estudos anteriores demonstraram elevado potencial citotóxico do derivado quinoxalínico PJOV56 contra células de câncer cólon, HCT-116. A fim de investigar a ação desse composto, este trabalho objetivou esclarecer a participação da senescência e da autofagia na inibição do crescimento celular induzido por PJOV56. A análise de reversão do potencial proliferativo após interrupção do tratamento com 3 µM mostrou efeito antiproliferativo persistente pelos ensaios do Azul de Tripan e monitoramento de proliferação em tempo real (XCELLigence), enquanto que a avaliação por MTT e por citometria de fluxo indicaram recuperação do crescimento. O tratamento com 3 µM e 6 µM também promoveu mudanças na distribuição do ciclo celular, com acúmulo de células em S e em G0/G1, respectivamente. Por sua vez, a análise por microscopia óptica (hematoxilina/eosina) revelou que o tratamento ocasionou aumento do volume celular e da quantidade e tamanho de vesículas no citoplasma das células, peculiaridades comuns aos fenótipos autofágico e senescente, enquanto que a análise por microscopia confocal mostrou intensificação da pigmentação das células com laranja de acridina, indicando, assim, a provável ocorrência de autofagia. Por outro lado, o aumento da expressão de SA-β-galactosidase e da expressão dos genes LAMA5 e THBS1, mesmo após 144h de suspensão do tratamento, corroboram fortemente a evidência de estado de senescência após contato inicial com o composto. Adicionalmente, a análise por Western Blot indicou discreto aumento de Beclina-1 e o gradual ganho, concentração-dependente, na expressão de LC3-II, indicando autofagia. Desse modo, possivelmente o composto PJOV56 induz senescência como principal mecanismo antiproliferativo da molécula, nas concentrações e tempos testados, sobre a linhagem de câncer de cólon HCT-116 enquanto que a autofagia parece atuar como mecanismo efetor de morte das células cancerígenas em senescência, sugerindo PJOV56 como um novo protótipo com potencial anticâncer.
Abstract: Once the current cancer chemotherapy presents inconveniences such as invasiveness, side effects and non-responsiveness the continuous search for new antitumor agents remains as a crucial concern. In order to investigate the action mode of a previously reported cytotoxic quinoxaline derivative, PJOV56, this work aimed to elucidate the cellular senescence and autophagy occurence PJOV56-induced on colon cancer cells, HCT-116. The recovery analysis after PJOV56 3.0 μM treatment withdrawal resulted in persistent antiproliferative effect, showed by Trypan Blue and Real-time proliferation monitoring (XCELLigence) results, while the MTT and cell counting by flow cytometry results indicated recovered cell growth. The compound also promoted changes in cell cycle distribution causing cell accumulation in the S and G0/G1 phases at concentrations of 3.0 and 6.0 μM, respectively. Optical microscopy analysis revealed that the treatment caused increased cell volume and extensive cytoplasmic vacuolization, both common peculiarities to autophagic and senescent phenotypes. Moreover, acridine orange staining assessed by confocal microscopy was elevated, thus indicating autophagy activaction. Additionally the cells showed augmented expression of SA-β-galactosidasis and increased expression levels of LAMA5 and THBS1 genes even after 144 hours of treatment removal, strongly supportting the evidence of senescence after initial contact with the compound. Western Blot analysis indicated a slight increase of Beclin-1 and a gradual elevation in LC3-II levels in a dose-dependent manner, suggesting autophagy. Thus, cellular senescence PJOV56-induced possibly act as the main antiproliferative mechanism of the molecule, at the concentrations and times tested on HCT-116 cells. It is likely that autophagy may act as an effector mechanism of clearance of cancer senescent cells, and may be considered as a new potential anticancer prototype.
