Abstract: Our objective was to evaluate the role of soluble guanylate cyclase(sGC)activation in the gastro-protective effect of the HO-1/CO pathway against alendronate-induced gastric damage in rats. Rats were pretreated,once daily for 4 days,with saline,hemin(HO-1inducer),ordimanganese decacarbonyl (DMDC,COdonor). Another group received zinc protoporphyrinIX(ZnPPIX,HO-1antagonist)1h before hemin treatmen torsGCinhibitor(ODQ)30min before hemin and DMDC treatment. After 30 min,gastric damage was induced by alendronate(30mg/kg)by gavage.On the last day of treatment, 4 h after alendronate administration,the animals were killed.Gastric lesions were measure dusing a computer planimetry program,and gastric corpus pieces were assayed formal ond ialdehyde(MDA), glutathione(GSH),pro-inflammatorycytokines(tumornecrosisfactor[TNF]-a and interleukin[IL]-1b), myeloperoxidase(MPO),orbilirubin.Anothergroupwasusedtomeasuregastricmucus.HO-1 expressionwasdetermined after salineoralendronate administration by immunohistochemistry. Alendronate in duced gastric damage,produced neutrophil accumulation,in creased MDA levels and MPO activity,and reduced GSH and mucusin the gastrict issue.Alendronate also in creased HO-1 immuno reactivity and the level of bili rubinin gastric mucosa.Pretreatment with heminor DMDC reduced neutrophil infiltration and TNF-a, IL-1b, and MDA formation,and in creased the levels of GSH and mucus in the gastrict issue.ODQ completel yabolished the gastroprotective effectof he minand DMD Candin creased alendronate gastric damage.Our results suggest that the HO-1/CO path way plays a protectiverole against alendronate-induced gastric damage through mechanisms that can be dependentons GC activation.
